چکیده

Rapid analyte measurement platform (RAMPtrade mark) fluorogenic hand-held immunoassays (HHAs) were evaluated for inclusivity/sensitivity, exclusivity/specificity, sample matrix effects, ruggedness/stability, and reproducibility in detection of ricin (RCA(60)), a potential biological threat agent. The limit of detection of HHAs for RCA(60) was 14 ng/mL or approximately 140 pg/test. HHAs were inclusive in detection of ricin RCA(60), RCA(120), ricin A chain, and ricin B chain and exclusive in discrimination of RCA(60) from other toxins. None of the sample matrices tested affected assay performance. RAMPtrade mark ratios were tolerant of increases in sample volume, however, a decrease in sample volume of 25% resulted in significantly increased readings. RAMPtrade mark readings were highly reproducible lot-to-lot, cartridge-to-cartridge, day-to-day, and reader-to-reader.

A fluorogenic heterogeneous immunoassay for cardiac muscle troponin cTnI on a digital microfluidic device.

We describe a fluorogenic two-site noncompetitive heterogeneous immunoassay with magnetic beads on a low-voltage digital microfluidic platform using closed electrowetting-on-dielectric (EWOD). All the steps of an enzyme-linked immunosorbent assay (ELISA) were performed on the device using 9H-(1, 3-dichloro-9, 9-dimethylacridin-2-one-7-yl) phosphate as the fluorogenic substrate for the enzyme al...

Evaluation of a rapid analyte measurement platform for West Nile virus detection based on United States mosquito control programs.

The rapid analyte measurement platform (RAMP) system is an immunoassay test for West Nile virus (WNV) detection. Although reverse transcriptase-polymerase chain reaction (RT-PCR) methodology has been regarded as the gold standard for confirming WNV presence, usage of RAMP testing kits has increased in the past years. We collected RAMP test result data that were subsequently confirmed with RT-PC...

A glyco-chip for the detection of ricin by an automated chemiluminescence read-out system.

The plant toxin ricin is a lectin that binds to D-galactose or lactose moieties by multivalent interactions. In the present work, this avidity was used to develop a novel sandwich glyco-immunoassay using a carbohydrate microarray. For realization, 6-azidohexyl-lactose was immobilized on an alkyne silane surface by Cu(I) catalyzed click chemistry. This procedure is fast, and prevents any nonspec...

Investigating a Functionalized Terthiophene Surface for the Detection of Progesterone Using Surface Plasmon Resonance

Herein we report a rapid and sensitive surface Plasmon resonance (SPR) biosensor to detect progesterone (P4), a model analyte of small molecules. A terthiophene scaffold with an oligoethylene glycol (OEG) linker is used as an anchor for the P4-ovalbumin (OVA) immobilization on gold to form a stable, regenerable SPR surface. Subsequently, inhibition SPR immunoassays based on this terthiophene/OV...

Rapid, automated, parallel quantitative immunoassays using highly integrated microfluidics and AlphaLISA

Immunoassays represent one of the most popular analytical methods for detection and quantification of biomolecules. However, conventional immunoassays such as ELISA and flow cytometry, even though providing high sensitivity and specificity and multiplexing capability, can be labor-intensive and prone to human error, making them unsuitable for standardized clinical diagnoses. Using a commerciali...
